Один из основных противотуберкулезных препаратов – рифампицин (РИФ) – является субстратом для P-gp, что снижает его эффективность. В статье представлены данные об активности P-gp в провоспалительных макрофагах человека и проведена оценка влияния на его активность новой формы РИФ, инкапсулированного в наночастицы полимера молочной кислоты, разрешенного для медицинского применения. Показано, что макрофаги ТНР-1, в отличие от исходных клеток, характеризуются возрастанием функциональной активности P-gp в процессе дифференцировки, а также сохранением жизнеспособности в присутствии РИФ, что связано с активацией P-gp. Новая форма РИФ является нетоксичной для моноцитов и макрофагов ТНР-1 в сравнении с традиционной формой. При использовании инкапсулированной формы РИФ происходят активация процесса эндоцитоза/фагоцитоза в макрофагах и снижение функциональной активности P-gp. Полученные данные демонстрируют, что создание инкапсулированных противотуберкулезных препаратов – активаторов фагоцитоза, направленно снижающих активность P-gp и таким образом влияющих на функциональные особенности макрофагов человека, представляется перспективным и актуальным направлением для повышения эффективности действия противотуберкулезных препаратов.</p></abstract><trans-abstract xml:lang="en"><p>Currently, the problem of effectiveness reduction of the anti-tuberculosis drugs is associated not only with the development of drug resistance by the pathogen, but also with the activity of the multidrug resistance protein P-gp (P-glycoprotein) of macroorganism cells. Rifampicin, the main anti-tuberculosis drug, is a substrate for P-gp. The article presents data on the activity of P-gp in pro-inflammatory human macrophages and assesses the effect of a new form of the anti-tuberculosis drug – rifampicin-loaded lactic acid polymer nanoparticles – on it. Scanning electron microscopy, laser confocal microscopy, MTT test and flow cytometry methods were used. It was shown that the new form of rifampicin is non-toxic for human monocytes and macrophages of the THP-1 cell line. In addition, it activates the processes of endocytosis/ phagocytosis and reduces the functional activity of P-gp.
