В настоящей работе проведена оценка влияния факторов, определяющих продукцию ВКМ, на геометрические параметры, структуру и продукцию коллагеновых волокон МСК из ткани пупочного канатика человека (МСК-ТПК). Исследовано влияние покрытия культуральных поверхностей желатином, продолжительности культивирования, концентрации фетальной бычьей сыворотки, а также добавок, стимулирующих продукцию ВКМ, – 2-фосфо-L-аскорбата натрия и каррагинана – на структуру коллагенового каркаса ВКМ и относительное содержание коллагеновых белков. Оптимальные условия включали культивирование МСК-ТПК на желатиновом покрытии в течение 11 сут в среде α-MEM с 10% фетальной бычьей сывороткой и добавлением аскорбата (50 мкМ). Эти параметры обеспечивали высокую адгезию, сохранность и отсутствие отслоений клеточного пласта, а также формирование коллагеновой сети ВКМ. Морфометрический анализ показал, что коллагеновый каркас ВКМ, полученный в таких условиях, характеризуется равномерным распределением, увеличением сонаправленности и числа пересечений волокон, сниженной лакунарностью и выраженной анизотропностью структуры. Относительное содержание коллагеновых белков было значимо выше, чем при других вариантах культивирования. Полученные данные позволяют рекомендовать разработанный протокол как эффективную и воспроизводимую платформу для получения биомиметических ВКМ, пригодных для задач клеточной инженерии и тканевой терапии.</p></abstract><trans-abstract xml:lang="en"><p>The extracellular matrix (ECM) synthesized by multipotent mesenchymal stromal cells (MSCs) plays a critical role in regulating the cellular microenvironment and is widely used in the development of biomaterials for regenerative medicine. The present study demonstrates the influence of cell culture conditions on ECM collagenous compartments secreted by umbilical cord derived MSCs (MSCs-UC) including the geometric parameters, structure complexity, and production of collagenous proteins. The impacts of gelatin coating, cell culture duration, fetal bovine serum concentration, and ECM-stimulating supplements (sodium 2-phospho-L-ascorbate and carrageenan) were examined. The optimal protocol included cultivation of MSCs-UC on gelatin-coated plated for 11 days in α-MEM supplemented with 10% FBS and 50 μM sodium ascorbate. These conditions supported high cell adhesion, structural integrity of the cell layer, and formation of a collagen-rich ECM network. Image analysis revealed that the collagenous fibers under these conditions exhibited a uniform distribution, enhanced fiber alignment and number of branching points, reduced lacunarity, and pronounced anisotropy. The relative collagen content under these conditions was significantly higher than in other culture setups.
