Ранее было показано, что Nhp6 входит в состав нескольких комплексов (в том числе комплекса FACT), присутствует на многих дрожжевых промоторах и транскрибируемых участках генов in vivo и участвует в процессе дестабилизации структуры нуклеосом in vitro. В нашей лаборатории был изучен фактор FACT и показана его роль в трaнскрипции хроматина эукариотической РНК-полимеразой 2 in vitro, однако роль белка Nhp6 в транскрипции ранее не была исследована. В данной работе мы изучили влияние этого белка на транскрипцию нуклеосом эукариотической РНК-полимеразой 2 и показали, что он увеличивает эффективность транскрипции в нескольких положениях на нуклеосомной ДНК, в первую очередь – участка в положении +(11–17) в нуклеосоме. Предложена модель действия Nhp6, предполагающая стабилизацию транзитного отворачивания ДНК от октамера гистонов в процессе транскрипции хроматина.</p></abstract><trans-abstract xml:lang="en"><p>Nhp6 is a small yeast protein, which binds DNA nonspecifically. It was shown that Nhp6 is a part of several protein complexes (including FACT complex) and is presented on many yeast promoters and transcribed regions of genes in vivo. It also participates in the process of destabilizing the structure of nucleosomes in vitro. In our laboratory, we studied FACT complex and showed its role in the transcription of eukaryotic RNA-polymerase 2 in vitro, but the role of the Nhp6 protein in transcription has not been studied previously. In this paper, we describe the effect of Nhp6 protein on transcription through a nucleosome by eukaryotic RNA-polymerase 2 and show that Nhp6 protein increases the transcription efficiency at several positions on the nucleosomal DNA, primarily the transcription at the positions +(11–17) in the nucleosome.
