CHARACTERISTICS OF REGULATION OF THE ACTIVITY OF GLUCOKINASE FROM RAT LIVER

Abstract

A simple method for purification of glucokinase from soluble fraction of rat liver has been developed which allows to obtain enzyme preparation with high specific activity and native cooperative characteristics practically lacking of contamination of other molecular forms of hexokinase. The dependence of enzyme activity upon glucose concentration shows pronounced sigmoid behavior: at 25°С and 2°С the Hillґs coefficients on whole range of substrate concentration are equal to 1,76 and 1,35, and S0,5 values for glucose are equal 6,9 mM and 20 mM, respectively. The role of kinetic mechanism of the reaction in the regulation of glucokinase activity in liver is discussed.
